The glucagon receptor is a cell-surface protein found mainly on liver cells, belonging to a family of receptors that transmit signals from hormones into biochemical instructions inside the cell. Its natural ligand, glucagon, is a peptide hormone released by the pancreas when blood glucose falls too low. When glucagon binds and activates its receptor, the liver mobilizes stored sugar and releases it into the bloodstream, counteracting hypoglycemia (dangerously low blood glucose). Rescue therapies mimic this hormone directly using glucagon-receptor agonist peptides administered as an injection or, for some agents, as a nasal spray, allowing rapid activation of the receptor when hypoglycemia occurs outside a clinical setting. This mechanism is centrally relevant to diabetes management, where insulin dosing carries the risk of over-lowering blood glucose, sometimes to a degree that impairs consciousness and prevents self-treatment with oral carbohydrate. Because such episodes require intervention from another person, the ability to administer a ready-to-use glucagon agonist without reconstitution or intravenous access is a key practical rationale for these products, extending the underlying pancreas-liver biology into the design of emergency-use pharmaceuticals for a life-threatening but reversible metabolic emergency.
