The GABA transporter is a membrane protein located on presynaptic nerve terminals and surrounding glial cells that removes the neurotransmitter gamma-aminobutyric acid (GABA) from the synaptic space after it has been released. GABA is the principal inhibitory neurotransmitter in the central nervous system, meaning its binding to postsynaptic receptors reduces the likelihood that a neuron will fire. By blocking the transporter, a drug can prolong the presence of GABA in and around the synapse, extending its inhibitory signaling on postsynaptic receptors. This approach—reuptake inhibition rather than direct receptor activation—aims to amplify the brain's own inhibitory tone using the timing and location of naturally released GABA, rather than substituting an external agonist. This mechanism is relevant wherever excessive or poorly controlled neuronal excitability is part of a disease process, most notably in epilepsy, where enhanced inhibition can raise the seizure threshold.
