The D1 receptor is a subtype of dopamine receptor belonging to the G-protein-coupled receptor family. When dopamine binds it, the receptor activates an intracellular enzyme called adenylyl cyclase, raising levels of the signaling molecule cyclic AMP and triggering downstream effects that influence neuronal excitability, synaptic plasticity, and cognitive processes such as working memory and executive function. Because D1 signaling in cortical and striatal circuits is implicated in cognitive and motor symptoms across several neuropsychiatric and neurodegenerative conditions, restoring or fine-tuning this pathway is of long-standing pharmacological interest. Rather than directly activating the receptor, a positive allosteric modulator binds a separate site on the receptor and amplifies the response to dopamine that is already present, without acting as a standalone trigger. This approach aims to preserve the natural timing and location of dopamine release, potentially offering a more physiological way to enhance signaling compared with direct agonists. This mechanism is broadly relevant to disorders in which dopaminergic transmission in the prefrontal cortex or striatum is diminished, including cognitive impairment associated with schizophrenia and Parkinson's disease, where restoring normal signaling dynamics rather than overriding them may be therapeutically advantageous.
