CCR2 chemokine receptor antagonist

CCR2 is a cell-surface receptor belonging to the chemokine receptor family, a group of G protein-coupled receptors that direct the movement of immune cells through the body. Its principal ligand is CCL2 (also called monocyte chemoattractant protein-1), a small signaling protein released by tissues undergoing injury, infection, or metabolic stress. When CCL2 binds CCR2 on circulating monocytes, it guides these cells out of the bloodstream and into the affected tissue, where they can mature into macrophages and contribute to inflammation, tissue remodeling, or fibrosis. Blocking CCR2 with an antagonist interrupts this recruitment step, reducing the influx of monocytes and macrophages into inflamed or damaged tissue. This rationale has made CCR2 a target of interest across a range of conditions where excessive macrophage accumulation drives disease, including chronic inflammatory disorders, fibrotic diseases of the liver and other organs, atherosclerosis, and certain cancers where tumor-associated macrophages support tumor growth and immune evasion. Because CCR2 signaling is one of several overlapping chemokine pathways that control immune cell trafficking, its modulation is generally viewed as a way to dampen a specific arm of the inflammatory response rather than to suppress immunity broadly.
