The aryl hydrocarbon receptor (AhR) is a protein inside cells that, once activated by a binding molecule (a ligand), moves into the nucleus and switches on specific genes. It responds to compounds from the environment, diet, and skin microbes, and its natural role includes helping cells process foreign chemicals and maintain tissue balance. In skin biology, AhR activation promotes genes involved in barrier formation and shifts immune signaling away from inflammatory pathways associated with excess interleukin-17 and interleukin-22 production, favoring a more regulated immune state. Because of this dual action on skin structure and local immune tone, AhR agonists are of interest for chronic inflammatory skin conditions such as psoriasis and atopic dermatitis, where both barrier dysfunction and inflammatory T-cell activity contribute to disease. Modulating this single receptor offers a way to address two related problems—impaired skin barrier and dysregulated inflammation—through one convergent pathway, rather than requiring separate agents for each. The broader relevance of AhR extends to research in autoimmune and inflammatory disease generally, given its role in shaping T-cell differentiation, though its most established therapeutic application is in dermatologic inflammation.
