Adiponectin receptors, AdipoR1 and AdipoR2, are cell-surface proteins that respond to adiponectin, a hormone secreted by fat tissue that helps regulate how the body handles glucose and fatty acids. Unlike most metabolic hormones, adiponectin levels tend to fall as fat mass and insulin resistance increase, so its signaling is generally considered protective rather than harmful. AdipoR1 is most abundant in skeletal muscle and favors pathways that promote fatty-acid burning and glucose uptake, while AdipoR2 predominates in liver and favors pathways that reduce fat accumulation and inflammation there. Because native adiponectin is a large, complex protein that is difficult to administer as a medicine, synthetic agonists are designed to engage these receptors directly and reproduce their downstream metabolic effects with a simpler molecule. This mechanism is broadly relevant to conditions involving insulin resistance, type 2 diabetes, fatty liver disease, and other metabolic disorders, where restoring or amplifying adiponectin-receptor signaling could improve energy metabolism and reduce tissue inflammation without necessarily targeting a single organ in isolation.
