This was a large, randomized, triple-blind, placebo-controlled trial with adequate sample size and hard clinical/functional/radiographic primary endpoints (ACR20 response, HAQ-DI, and van der Heijde-modified Total Sharp Score). In the confirmatory Part B, both sarilumab 150 mg and 200 mg every 2 weeks met all three primary endpoints with highly significant differences from placebo (p<0.0001), and Part A also showed significant ACR20 responses for several sarilumab doses. The positive, well-designed confirmatory results support strong evidence for sarilumab.

This randomized, double-blind, active-comparator trial of 340 participants met its primary endpoint: about half of tocilizumab-treated patients achieved an ACR20 response at 24 weeks, compared with a quarter on methotrexate alone, a highly statistically significant difference for both the combination and monotherapy arms.
