Head-to-head evidence

Panobinostat vs Purinostat Mesylate

Same mechanism, same race: design-graded trial evidence for both assets, with the grading rationale shown. Grades describe the quality of the evidence, never the merits of either security.

Panobinostat

Marketed by Novartis

Approved in Multiple Myelomasmall molecule (pan-HDAC inhibitor)
Partial signal

NCT01023308 · Phase 3 · completed

This was a randomized, double-blind, placebo-controlled Phase 3 trial with a large sample (768 patients) and a hard clinical endpoint (progression-free survival), which are all strong design features. However, the posted results show only the median PFS for each arm (12.0 vs 8.8 months) without a stated p-value, hazard ratio, or confidence interval for the treatment comparison, so whether this difference was formally statistically significant cannot be confirmed from the record, capping this at a more limited grade despite the otherwise excellent design.
