This Phase 3 trial randomly assigned adults with moderate-to-severe atopic dermatitis to manfidokimab (AK120) or placebo, both delivered by subcutaneous injection, under quadruple-blind conditions, with the proportion of patients achieving EASI-75 (75% improvement in eczema severity) at week 16 as the primary endpoint. No results have been posted, so this remains a well-designed, adequately sized, blinded, placebo-controlled trial whose findings are not yet known.

The study was randomized, quadruple-blind, placebo-controlled, and included 330 participants, with a clinical primary endpoint based on validated Investigator Global Assessment. Although the reported primary response favored rademikibart (29.0% vs 5.9%), the registry does not state whether the primary comparison was statistically significant, so the result remains unclear.
