The trial randomly assigned 344 participants to inhaled loxapine 5 mg, loxapine 10 mg, or placebo and used quadruple blinding of participants, care providers, investigators, and outcome assessors. Its primary clinical agitation measure was significantly improved with both loxapine doses compared with placebo (p=0.0004 and p<0.0001).

The 675-participant study was randomized, quadruple-blind, and placebo-controlled, with approximately 165 to 169 participants per blinded treatment group. Its primary clinical endpoint, change in PANSS total score at week 8, was significantly better with each TV-44749 dose than with placebo (all adjusted p<0.0001), supporting a strong evidence rating for the blinded efficacy comparison.
