In this 195-participant randomized, double-blind, placebo-controlled trial, the primary clinical endpoint was met: pooled CIN-107 reduced seated systolic blood pressure by 15.22 mmHg versus 7.15 mmHg with placebo, a between-group difference of 8.08 mmHg (p=0.003). The trial had a substantial sample size and the primary comparison was statistically significant in the favorable direction.

This was a randomized, quadruple-blind, placebo-controlled phase 2 trial in 586 patients with chronic kidney disease, comparing three doses of vicadrostat (3, 10, and 20 mg) against placebo, all on a background of empagliflozin or placebo. The primary endpoint was the change in urine albumin-to-creatinine ratio (UACR), a physiological measure that defines and stages CKD, at 14 weeks; the 10 mg and 20 mg doses achieved significantly larger reductions than placebo (p<0.0001) and the dose-response analysis was significant, so the primary endpoint was met. The 3 mg dose was only borderline (p=0.0499), making the overall result favorable but clearly dose-dependent.
