Lorlatinib

Marketed by Pfizer

Approved in Non-Small Cell Lung Cancersmall molecule
Weak signal

NCT01970865 · Phase 1/2 · completed

This was an open-label, non-randomized, single-arm study with no concurrent control group, despite enrolling 364 patients across dose-escalation and expansion cohorts. The primary phase 2 endpoint was objective response rate by RECIST v1.1, a clinical response measure; response rates were reported descriptively, but there was no randomized comparator or blinding, so the results cannot establish comparative efficacy and are best interpreted as uncontrolled evidence.

Next expected readout: December 2026 · NCT05059522 (registry estimate)

XZP-3621

Developed by Xuanzhu Biopharmaceutical

Phase 3 in Non-Small Cell Lung Cancersmall molecule
Partial signal

NCT05204628 · Phase 3 · status unknown

Patients were randomly assigned to XZP-3621 or crizotinib, but the study is open-label rather than blinded, which is a weaker design even though it measures a hard clinical outcome, progression-free survival, against an active standard-of-care comparator. Results have not been posted yet.
