Head-to-head evidence

Glecirasib vs Sotorasib

Same mechanism, same race: design-graded trial evidence for both assets, with the grading rationale shown. Grades describe the quality of the evidence, never the merits of either security.

Glecirasib

Developed by Jacobio Pharmaceuticals

Phase 3 in Non-Small Cell Lung Cancersmall molecule
Weak signal

NCT05002270 · Phase 1/2 · completed

The study was a small, non-randomized, open-label trial without a comparator, evaluating safety, tumor response, and duration of response. Because results are not posted and the design cannot provide a controlled comparison, it offers limited evidence for glecirasib.

Sotorasib

Marketed by Amgen and partners

Approved in Non-Small Cell Lung Cancersmall molecule
Partial signal

NCT04303780 · Phase 3 · completed

The 345 participants were randomized 1:1 to sotorasib or active-comparator docetaxel, but treatment assignment was not masked. The primary clinical endpoint, progression-free survival assessed by blinded independent central review, was met in favor of sotorasib, with median progression-free survival of 5.62 versus 4.47 months and a hazard ratio of 0.663 (p=0.002).

Next expected readout: October 2026 · NCT06333951 (registry estimate)
