Enlicitide

Marketed by Merck & Co.

Phase 3 in Atherosclerotic Cardiovascular Diseasepeptide
Reliable signal

NCT05952856 · Phase 3 · completed

This was a randomized, triple-blind, placebo-controlled Phase 3 trial in 2,909 adults with hypercholesterolemia. The primary endpoint was the mean percent change in LDL-C from baseline to Week 24, a standard regulator-accepted efficacy measure for lipid-lowering therapy. The active arm achieved a significantly greater LDL-C reduction than placebo (-57.1% vs +3.0%, p<0.001), so the trial met its primary endpoint with a large effect size and low risk of bias.

Evolocumab · Repatha

Marketed by Amgen

Approved in Atherosclerotic Cardiovascular Diseasemonoclonal antibody
Partial signal

NCT01516879 · Phase 3 · completed

The study randomly assigned 905 participants to quadruple-blinded monthly evolocumab or placebo, with both groups continuing background lipid-lowering therapy. The primary endpoint was met, with a statistically significant difference in LDL-C change at week 52, but LDL-C is a surrogate laboratory endpoint rather than a clinical outcome.

Next expected readout: November 2026 · NCT06496243 (registry estimate)
