Reliable signal

NCT00708162 · Phase 3 · completed

This was a randomized, triple-blind (participant, care provider, investigator), double-dummy, active-controlled phase 3 trial in 724 treatment-experienced HIV patients, comparing elvitegravir with raltegravir, both given with an optimized background regimen. The primary endpoint, confirmed HIV-1 RNA <50 copies/mL at week 48, was met as a non-inferiority result: 59.0% with elvitegravir vs 57.8% with raltegravir, with the 95% confidence interval for the difference (-6.0 to 8.2) falling entirely within the prespecified 10% margin. The trial was adequately powered (planned 700 participants, 85% power for non-inferiority).

Reliable signal

NCT00293254 · Phase 3 · completed

This was a randomized, double-blind, placebo-controlled trial with 351 patients, using HIV-1 RNA suppression - a regulator-accepted, disease-defining measure - as the primary endpoint. The raltegravir arm showed clearly higher suppression rates than placebo at both Week 16 and Week 48, with confidence intervals that do not overlap, indicating a statistically robust and clinically meaningful benefit.
