Head-to-head evidence

dupilumab vs Manfidokimab

Same mechanism, same race: design-graded trial evidence for both assets, with the grading rationale shown. Grades describe the quality of the evidence, never the merits of either security.

Atopic Dermatitis

Shared mechanism: IL-4R-alpha · full Atopic Dermatitis pipeline

dupilumab · Dupixent

Marketed by Regeneron Pharmaceuticals and partners

Approved in Atopic Dermatitismonoclonal antibody
Partial signal

NCT01949311 · Phase 3 · completed

This was an explicitly described open-label extension enrolling participants from prior dupilumab atopic dermatitis trials, including the SOLO 1 and SOLO 2 programs, with 2,677 participants receiving dupilumab in a single group. The primary outcomes were safety events rather than a comparative efficacy endpoint, and there was no randomization, blinding, or control arm; therefore, the findings provide limited evidence despite the large enrollment.

Manfidokimab

Developed by Akeso

Phase 3 in Atopic Dermatitismonoclonal antibody
Partial signal

NCT06383468 · Phase 3 · status unknown

This Phase 3 trial randomly assigned adults with moderate-to-severe atopic dermatitis to manfidokimab (AK120) or placebo, both delivered by subcutaneous injection, under quadruple-blind conditions, with the proportion of patients achieving EASI-75 (75% improvement in eczema severity) at week 16 as the primary endpoint. No results have been posted, so this remains a well-designed, adequately sized, blinded, placebo-controlled trial whose findings are not yet known.

Asthma

Shared mechanism: IL-4R-alpha · full Asthma pipeline

dupilumab · Dupixent

Marketed by Regeneron Pharmaceuticals and partners

Approved in Asthmamonoclonal antibody
Partial signal

NCT03782532 · Phase 3 · completed

The study was randomized, triple-blind, and placebo-controlled with 486 participants, which is a substantial and generally rigorous design. However, the primary endpoint was a lung-function measure rather than a hard clinical outcome, and the registry does not report whether the comparison was statistically significant.

Next expected readout: November 2026 · NCT05720325 (registry estimate)
