This was a randomized, quadruple-blind, active-controlled phase 3 trial in 833 participants comparing dolutegravir plus abacavir/lamivudine to Atripla. The primary endpoint of virologic suppression (<50 copies/mL) at week 48 was a hard clinical measure, and the comparison was statistically significant in favor of dolutegravir (88% vs 81%, p=0.003), with the confidence interval excluding zero. The trial was adequately powered and the primary outcome was met, supporting a strong evidence grade.

This was a randomized, triple-blind (participant, care provider, investigator), double-dummy, active-controlled phase 3 trial in 724 treatment-experienced HIV patients, comparing elvitegravir with raltegravir, both given with an optimized background regimen. The primary endpoint, confirmed HIV-1 RNA <50 copies/mL at week 48, was met as a non-inferiority result: 59.0% with elvitegravir vs 57.8% with raltegravir, with the 95% confidence interval for the difference (-6.0 to 8.2) falling entirely within the prespecified 10% margin. The trial was adequately powered (planned 700 participants, 85% power for non-inferiority).
