Dimethyl fumarate (Tecfidera)
Marketed by Biogen
NCT00451451 · Phase 3 · completed
This was a large (n=1417), randomized, double-blind, placebo-controlled Phase 3 trial with a hard clinical primary endpoint (annualized relapse rate, independently adjudicated). Both BG00012 dosing arms showed clearly separated confidence intervals from placebo on this primary endpoint (0.224 and 0.198 relapses/year vs 0.401 for placebo), indicating the drug met its primary comparison in the favorable direction, satisfying all the elements of well-designed, positive Phase 3 evidence.

Diroximel fumarate · Vumerity
Marketed by Biogen
NCT05083923 · Phase 3 · completed
The study was a single-group, open-label trial with 136 participants enrolled and no placebo or active comparator. Its primary outcomes were safety measures rather than a comparative clinical efficacy endpoint, so there was no formal primary treatment comparison to meet or miss; the reported safety findings are descriptive only.
Next expected readout: July 2034 · NCT07483632 (registry estimate)
