The initial 4-week comparison was randomized, triple-masked, placebo-controlled, and included 730 participants; the primary sitting systolic blood pressure endpoint was statistically significant for both aprocitentan doses. Blood pressure is the regulator-accepted primary efficacy measure for antihypertensive therapy, and the prespecified comparison being met in the favorable direction supports strong evidence.

Because everyone in this study received bosentan, with the great majority having rolled over directly from the earlier randomized, placebo-controlled BENEFIT trial, there is no control group here and no statistical comparison between arms; it tracks how patients did over time on the drug rather than proving the drug outperforms placebo, though it extends the earlier controlled trial's population with longer-term functional and safety follow-up.
