This was a randomized, double-blind, placebo-controlled trial in 102 adults with obesity taking tirzepatide, and apitegromab significantly reduced lean-mass loss compared with placebo (p=0.001). The primary measure was change in lean body mass, a body-composition measure, rather than the total weight loss that defines an obesity treatment's efficacy, so this is supportive evidence of a muscle-preserving effect rather than a direct efficacy readout for obesity.

This randomized, double-blind, placebo-controlled trial enrolled 507 adults and used a hard clinical primary endpoint, change in body weight at Week 48. The primary endpoint comparison showed a statistically significant advantage for most active treatment combinations over placebo, including the highest-dose combination (bimagrumab 30 mg/kg + semaglutide 2.4 mg), but the 10 mg/kg bimagrumab monotherapy arm missed significance, yielding a mixed primary endpoint result. Because the primary endpoint was not uniformly met across the multiple prespecified comparisons, the overall evidence is moderate despite the strong design.
