Alimatravir

Developed by Merck & Co.

Phase 3 in HIVsmall molecule
Partial signal

NCT06045507 · Phase 2 · completed

The study randomized 352 participants to three MK-8527 dose groups or matched placebo and used double blinding, but Alimatravir was not studied and was not the comparator. Its primary endpoints were safety outcomes rather than efficacy, and the registry provides counts without statistical comparisons, so whether the primary comparisons were met cannot be determined.

Doravirine · Pifeltro

Marketed by Merck & Co.

Approved in HIVsmall molecule
Reliable signal

NCT02275780 · Phase 3 · completed

This was a randomized, double-blind, active-controlled Phase 3 trial with 769 enrolled adults (385 doravirine, 384 darunavir/ritonavir). The primary endpoint was virologic suppression (HIV-1 RNA <50 copies/mL) at Week 48, a hard clinical endpoint. The non-inferiority comparison was met: the difference was 3.9 percentage points (95% CI -1.6 to 9.4), with the lower bound above the prespecified -10% margin. The trial therefore provides strong evidence that doravirine is non-inferior to darunavir/ritonavir.
