Head-to-head evidence
Afatinib vs Pyrotinib
Same mechanism, same race: design-graded trial evidence for both assets, with the grading rationale shown. Grades describe the quality of the evidence, never the merits of either security.

Afatinib · Gilotrif
Marketed by Boehringer Ingelheim
NCT00949650 · Phase 3 · completed
The study randomly assigned 345 participants to afatinib or active chemotherapy, but it was open-label rather than blinded. Its hard clinical primary endpoint, centrally assessed progression-free survival, was statistically significantly better with afatinib, but the lack of blinding limits the strength of the evidence.

Pyrotinib
Developed by Jiangsu Hengrui Pharmaceuticals
NCT04447118 · Phase 3 · completed
The study randomly assigned 151 participants to pyrotinib or active treatment with docetaxel, with the primary endpoint assessed by an independent committee blinded to treatment assignment. Because results for the progression-free survival comparison have not been posted, the trial provides limited evidence at this time despite its randomized design.
Next expected readout: December 2026 · NCT05482568 (registry estimate)
