Abemaciclib · Verzenio

Marketed by Eli Lilly

Approved in HR+/HER2- Metastatic Breast Cancersmall molecule
Weak signal

NCT02102490 · Phase 2 · completed

This was a single-arm, open-label study of 132 participants with no placebo or active comparator and no randomization or blinding. Its primary endpoint was the clinical tumor-response rate, reported as 19.7%, but the uncontrolled design makes the result descriptive rather than comparative evidence.

Marketed by Novartis

Approved in HR+/HER2- Metastatic Breast Cancersmall molecule
Reliable signal

NCT02422615 · Phase 3 · completed

The study randomly assigned 726 participants to ribociclib plus fulvestrant or placebo plus fulvestrant and was quadruple-blinded for the primary comparison. Its radiographically assessed progression-free survival endpoint was significantly better with ribociclib; participants were later unblinded and offered crossover after a protocol amendment.
